微透析电化学检测在线联用实时分析烟碱依赖大鼠伏隔核内单胺化合物的变化

In Vivo Study on the Monoamine Compounds in Rat Nucleus Accumbens Exposed to Nicotine by Online Microdialysis Coupled with Electrochemical Detector

  • 摘要: 为探讨烟碱依赖的形成机理,采用微透析和高效液相色谱-库仑阵列电化学检测器在线联用方法测定了大鼠伏隔核内多巴胺(DA)、3,4-二羟基苯乙酸(DOPAC)、高香草酸(HVA)、5-羟色胺(5-HT)和5-羟吲哚乙酸(5-HIAA)的含量变化.以1.5 <i<μ</i<L/min灌流,每15 min收集1次透析液并自动注入梯度洗脱条件下的色谱进行检测.1.0 mg/kg烟碱暴露后,正常大鼠伏隔核内DA和5-HT的含量分别为基础水平的149%和134%,代谢产物DOPAC、HVA和5-HIAA的含量显著升高;烟碱依赖大鼠脑内5种单胺化合物的含量低于盐水对照组,1.0 mg/kg烟碱暴露后DA和5-HT的含量分别为基础水平的125%和119%,3.0 mg/kg烟碱暴露使DA和5-HT的释放增加至基础水平的147%和135%.结果表明,与正常大鼠相比,烟碱依赖发生后,更高剂量的烟碱方能激发同等水平的单胺神经递质释放及代谢变化特征.本研究为探讨烟碱依赖的形成机理提供了新的数据参考.

     

    Abstract: To investigate potential mechanism of nicotine dependence, online microdialysis sampling coupled with electrochemical detector was applied to study the contents of dopamine (DA), 3,4-dihydroxyphenyl acetic acid (DOPAC), homovanillic acid (HVA), serotonin (5-HT) and 5-hydroxylindole acetic acid (5-HIAA) in the nucleus accumbens of alive rats exposed to nicotine.Dialysates were collected every 15 min at a flow rate of 1.5 <i<μ</i<L/min and separated immediately under the gradient elution condition.After nicotine administration (1.0 mg/kg, s.c.), the release of DA and 5-HT increased significantly, and the metabolites DOPAC, HVA and 5-HIAA were raised continuously.In nicotine-dependence rats, 3.0 mg/kg nicotine injection could stimulate the equivalent release of DA, 5-HT and their metabolites.The results provide an alternative approach and data reference for studying the mechanism of nicotine dependence.
