桦树皮提取物抑制血管生成作用的研究

Birch Bark Extracts Suppression of Proliferative Ability of the Human Umbilical Vein Endothelial Cells

  • 摘要: 研究桦树皮提取物对血管生成的抑制作用.应用MTT法检测桦树皮提取物对人脐静脉内皮细胞的增殖抑制作用,采用Realtime PCR方法检测桦树皮提取物对人脐静脉内皮细胞中VEGFmRNA表达的影响,应用ELISA测定细胞上清液中VEGF的分泌情况.实验表明,桦树皮提取物能显著抑制人脐静脉内皮细胞增殖,最高浓度150 μg/mL其抑制率达48.4%,能明显抑制人脐静脉内皮细胞内VEGFmRNA的表达及VEGF分泌.结果表明桦树皮提取物可通过抑制细胞内VEGF mRNA的表达及VEGF的分泌有效抑制人脐静脉内皮细胞的增殖.

     

    Abstract: To research birch bark extracts' proliferation inhibition in human umbilical vein endothelial cells. The proliferation inhibition of birch bark extracts (BBE) in human umbilical vein endothelial cells (HUVEC) was measured by MTT assays. Expression of VEGFmRNA was measured by PCR. VEGF was measured by ELISA. Cell growth was obviously suppressed in the BBE treated groups. 150 μg/mL group was significantly different from control group. Realtime PCR reveales that BBE treated groups cause the expression ratios of VEGFmRNA decreased from 1 to 0.125(75 μg/mL) and 1 to 0.05(150 μg/mL) in 48 h. ELISA showes that high concentration BBE treated group leads to significant reduction in the levels of VEGF. BBE inhibited the proliferation of HUVEC through inhibiting intracellular expression of VECF mRNA and secretion of VECF.
