Preliminary Screening of Peptide Biomarker for Early Diagnosis of Diabetes

  • 摘要: 基于蛋白质组学策略结合18O标记技术,分析人血清白蛋白糖基化程度的糖尿病生物标志物筛选模型.基于HPLC-ESI/TOF MS的检测方法,筛选对葡萄糖敏感的人血清白蛋白酶切肽段.建立糖尿病监测及早期预警肽类生物标志物筛选模型,初步筛选出11个潜在的糖尿病早期诊断生物标志物.

     

    Abstract: An in vitro glycation model was set up by glucose and human serum albumin (HSA) based on Maillard Reaction. HPLC-ESI TOF MS and 18O-labeling technique were employed to quantify glycation level of peptides cleaved from HSA. In order to screen glucose-sensitive peptides for being diabetic biomarker-candidates, the in vitro glycation model was set up and confirmed, and eleven biomarker-candidates were screened based on the model by HPLC-ESI TOF MS quantification. Eleven glucose-sensitive peptides were screened for being biomarker-candidates, which needs to be verified in clinical trials in the future.
