傣百解对模拟微重力下大鼠脾淋巴细胞功能的影响

Protective Effects of Daibaijie on Immune Function Under Simulated Microgravity Environment

  • 摘要: 探讨模拟航天飞行失重状态下大鼠免疫功能的变化和傣百解的免疫调节作用.将Wistar大鼠分为空白对照组、吊尾组、吊尾加傣百解低、中、高剂量组.连续吊尾21 d后比较各组大鼠脾淋巴细胞的增殖能力、免疫相关细胞因子分泌水平,以及脾淋巴细胞中NFκB和IκBα蛋白表达水平.结果显示与正常组比较,吊尾组大鼠脾淋巴细胞刀豆蛋白刺激后的增殖能力减弱,细胞因子IL-2、IL-6、IFN-γ释放减少,而且NFκB信号通路激活水平低于正常组.傣百解能够提高失重大鼠脾淋巴细胞的NFκB信号通路激活水平,诱导淋巴细胞增殖和释放细胞因子,具有改善免疫功能的作用.

     

    Abstract: Male Wistar rats were used to investigate the change of immunological function in rat models of simulation of space flight and the regulation effects of Daibaijie at SPF level. The rats assigned to five groups randomly: normal control group, tail suspended group, tail suspended group with low dose, tail suspended group with middle dose, and tail suspended group with high dose of Daibaijie extract. After 21 days, lymphocyte proliferation, IL-2, IL-6 and IFN-γ, NFκB and IκBα were compared in ConA challenged splenic lymphocyte. The results show that tail suspension can reduce the proliferation function of lymphocyte, decrease the production of IL-2, IL-6 and IFN-γ, and inhibite the activation of NFκB signal transduction pathway. Daibaijie extract can enhance activation of NFκB signal transduction pathway, and improve lymphocyte proliferation and release of cytokines, which indicates that Daibaijie could regulate cellular immunological function.
