细胞周期调控人神经母细胞瘤细胞辐射耐受性研究

The Relation Between the Cell Cycle and Radio-Resistance in Human Neuroblastoma SH-SY5Y Cells

  • 摘要: 为研究脑中肿瘤对辐射的敏感性,选择SH-SY5Y神经母细胞瘤为研究对象进行辐射,并对细胞增殖、细胞凋亡及细胞周期进行研究.采用γ-射线辐射,剂量分别为5,10及20 GY,通过细胞计数的实验方法,比较不同剂量辐射后的神经母细胞瘤细胞数目与不辐射细胞数目的差别,进而说明细胞的增殖情况;同时通过PI单染的实验方法比较细胞周期的不同;最后通过对凋亡蛋白caspase 3/7及caspase 8的活性进行测定比较辐射后凋亡的变化水平.发现在10及20 GY的辐射剂量辐射后,24 h时间点细胞的数量相比于5 GY及对照细胞发生了细胞数目的减少,说明在此剂量下细胞出现了死亡或细胞周期发生了明显的阻滞现象.对细胞周期的检测结果发现,5 GY细胞周期变化不明显,而10及20 GY的剂量辐射后细胞发生明显的阻滞现象.并且凋亡蛋白caspase 3/7及caspase 8蛋白活性随剂量的增加,时间点的延长而呈增加的趋势.研究结果表明,细胞辐射后存在细胞周期阻滞现象,且辐射剂量越大,细胞周期阻滞能力越强;伴随周期阻滞的缓解可出现细胞凋亡活性增加的趋势.

     

    Abstract: Aiming to research on the brain tumors irradiation sensitivity, the cell proliferation, apoptosis and cell cycle in the SH-SY5Y cell line following irradiation were measured. At different doses, SH-SY5Y cells damage were assessed by cell counting, PI staining and caspase 3/7 and caspase 8 activity. The cells damage is distinctive between 5 GY and 10 GY or 20 GY, and the high dose irradiation induces the cells number decreased and the apoptosis protein caspase 8 and caspase 3/7 activity increased. These results reveal the cell happen to be apoptosis and cell cycle arrest. The γ-ray irradiation results in cell cycle arrest and cell apoptosis, and cell damage level is associated with the irradiation dose. Although the cell cycle arrest inhibited apoptosis, the high dose still induces the cell apoptosis protein activity increased in cell cycle arrest process. Cycle arrest is proved to be very important in the radioprotective mechanism of SH-SY5Y cells.
