低速低压温度敏感硼酸亲和色谱制备

摘要: 利用原子自由基转移聚合原理,制备聚(N-异丙基丙烯酰胺-co-4-乙烯基苯硼酸)改性的温度敏感硼酸亲和色谱固定相,并对其进行傅里叶红外光谱,X射线光电子能谱等结构表征以及用热失重对其接枝率进行了测定.用HPLC研究了腺苷和脱氧腺苷的保留行为.并且应用于空间生命仪器搭载装置的分离装置中,在不同温度下对腺苷进行富集并对其收集液进行分析.结果表明,在水溶液为流动相的条件下,可以实现在低流速、低压的条件下对腺苷的富集,且回收率为85.4%.Abstract: Poly (N-isopropyl acrylamide-co-4-vinyl benzene boric acid) (P (NIPAAm-co-VPBA)) modified liquid chromatographic stationary phase on silica gel was prepared by atom transfer radical polymerization (ATRP) and was characterized by Fourier infrared spectrum, X-ray photoelectron spectroscopy. Thermogravimetric analysis was performed to determine its grafting ratio. The retention behavior of adenosine and deoxyadensine was studied by HPLC at low-pressure and low-flow velocity. The rate of recovery is 85.4%. The results indicate that the enrichment of adenosine can be realized in this self-made device, which can provide theoretical foundation and technical support for spatial device.
